Both players went round the Royal Johannesburg & Kensington Golf Club in 63 to lead by a shot from South African Bryce Easton with eight more players grouped together on six-under par.
Sterne, who finished runner-up to Stephen Gallagher in the Dubai Desert Classic last weekend, posted six birdies and an eagle in his round on the West Course, while Kieffer played the same course and shot eight birdies in his round.
Both will play the East Course on Friday and Sterne, who won the Joburg Open five years ago – one of five titles on the tour, told the European Tour’s website: “I had three good rounds on the East Course that year. If I can do that now, I’ll have a chance on Sunday.
“Some days it feels good and everything kind of happens. Golf is a strange game and you have to hit the right shots at the right time. I hit 17 greens today and was never in trouble.”
Easton struck a hole-in-one in his round on the East Course with his seven-under total setting a new course record.
